Objectives :

-To work out and implement a procedure starting from a modus operandi-To use and understand the different assay techniques and the measuring apparatus -To treat the results using the data-processing tool and to validate them with a critical spirit

Course Content :

1.Measuring of spectrophotometry

-Of emission

i.Atomic : Assay of Na+ and K+ in serum

ii.Molecular : initiation to fluorimetry; proportioning by fluorescence of the acetylsalicylic acid contained in an analgesic

-Of absorption,

i.Atomic : proportioning of Mg2+, of Fe3+

ii.Molecular : initiation to spectrophotometry of absorption, assay of inorganic phosphorus, of cholesterol, of seric iron and TIBC, of glucose

2.Qualitative and quantitative analysis of proteins  Enzymatic activity

Serum proteins separation by electrophoresis on agarose

Protein separation by electrophoresis on acrylamid gel (SDS-PAGE)

Serum proteins assay by the methods of Bradford, method to the biuret and of Lowry

Urinary proteins assay by turbidimetry

Enzymology : kinetic study and research of the activity of alkaline phosphatase, kinetic study and activity of the LDH

3.Statistical applications to proportionings

Serum proteins assay by the methods of Bradford : variance analysis with one dimension and mean comparison

Magnesium assay by atomic absorption : variance analysis with two dimensions, specificity (mean comparison), precision, correlation.

4.Chromatography

Qualitative and quantitative analysis of alcohols by GC

Qualitative and quantitative analysis of components from an analgesic by HPLC

Purification of the lactate deshydrogenase from the pig by chromatography by freezing-filtration (séphadex G25) and chromatography of ion exchange

5.Immunological proportionings

Radioimmunoasssay of estradiol

Proportioning by ELISA of the serum bovine albumin biotinyled Immunodetection of the human transferrine after electrophoresis in gel of acylamid and transfer of nitrocellulose membrane
